Runners in the Yokohama Marathon use a section of the Shuto Expressway Bayshore Line, with the full marathon course utilizing the elevated road between Sugita and Honmoku Pier. The event takes place in Yokohama in late October, offering a 42.195 km marathon, a two-person relay over the same distance, a 20.3 km Wangan Highway Run, a 7 km Minatomirai run, and a 3 km Enjoy Run. The marathon and relay begin at 8:30 AM and have a six-hour completion time, while the shorter events have their own time limits or, for the Enjoy Run, no chip timing. Awards are presented to the top eight men and women in the marathon and the Minatomirai 7 km, but not in the Wangan Highway Run or Enjoy Run.
The full course begins near Yokohama Landmark Tower, crosses Minatomirai Ohashi, passes the Central Wholesale Market, Pacifico Yokohama, the Red Brick Warehouse, Yokohama Customs, Yamashita Park, and Honmoku Citizens' Park, then turns around near Yokohama Southern Market before returning toward the waterfront finish at Rinko Park. The relay exchange point is near Isogo Sports Center, and the Wangan Highway Run starts near Southern Market Station before merging with the expressway and port-side finish route. The 7 km route stays within the Minatomirai and Red Brick Warehouse area, while the 3 km Enjoy Run connects Pacifico Yokohama, Yokohama Hammerhead, Megami Bridge, and Rinko Park. Participants include marathoners and relay teams, junior high students, elementary-school-and-guardian pairs, and wheelchair participants; racing wheelchairs are not permitted in the wheelchair Enjoy Run. A portion of each entry fee contributes to Yokohama Marathon's Green Action environmental program.
